Oral Mucositis Model - Acute Radiation

Provider: Founded in 1997, Biomodels is an AAALAC accredited lab in Watertown MA. Biomodels offers a variety of high quality animal models for compound screening.

Background: The acute radiation model or oral mucositis in hamsters is an accurate, efficient and cost-effective technique to provide a preliminary evaluation of anti-mucositis compounds. The course of mucositis in this model is well defined and results in peak scores approximately 14-16 days following radiation. The acute model has little systemic toxicity, resulting in few hamster deaths, thus permitting the use of smaller groups for initial efficacy studies. It has also been used to study specific mechanistic elements in the pathogenesis of mucositis. Molecules that show efficacy in the acute radiation model may be further evaluated in the more complex models of fractionated radiation, chemotherapy, or concomitant therapy.

Service Details: Thirty-two (32) male Syrian Golden Hamsters are randomly assigned to four (4) groups of eight (8) animals each. On study day 0, each hamster is given an acute radiation dose of 40 Gy directed to their left buccal cheek pouch. Test articles are administered topically, orally or by SC or IP injection daily on days -1 to 20. Animal condition is assessed daily and body weights were measured once daily from day -1 to day 28. Mucositis is evaluated on days 6, 8, 10, 12, 14, 16, 18, 20, 22, 24, 26 and 28. The duration and severity of mucositis are compared between treatment groups and the control group to determine the impact of the test article on the course of mucositis.

Deliverable: Full report with analysis of weight change, survival if appropriate and statistical analysis of mucositis, including raw data and photographs of mucositis. This Mucositis Scoring Scale provides guidelines on how we evaluate animals and effectiveness of testing articles. Report is reviewed & signed by an expert in the field of oral mucositis.
