Background: INDIGO Biosciences’ nuclear receptor assay products are cell-based, all-inclusive systems optimally configured for HTS applications. These assay systems feature engineered nuclear receptor-specific reporter cells prepared using INDIGO Biosciences’ proprietary CryoMite™ process. Reporter cells shipped on dry ice may be thawed and used immediately, or further stored at -80°C ready for on-demand use as consumable assay reagent. INDIGO Biosciences’ assays enable sensitive in vivo screening of test compounds to assess their agonist or antagonist activities against human nuclear receptors expressed within the cytoplasm and nuclear environments of fully functional, healthy, dividing mammalian cells.

Product Details: This LXRβ Reporter Assay System utilizes non-human mammalian cells engineered to express human NR1H2 protein, commonly referred to as LXRβ. Following ligand-activation, LXRβ acts to induce expression of the firefly luciferase reporter gene. Light emission from the luciferase reaction is quantified using a platereading luminometer, providing a direct correlation to the level of LXRβ activation. This assay includes a luciferase detection reagent formulated to provide stable light emission over at least 1 hour, thus eliminating the need to use a luminometer equipped with injectors, and dramatically reducing the overall read-time of the assay plate. The stable, extended luminescence signal enables convenient batch processing of assay plates, thereby making this assay system ideally suited for high-throughput screening applications

Deliverable: This assay kit is shipped on dry ice. Upon receipt, individual kit components may be stored at the temperatures indicated on their respective labels, and below. To ensure maximal viability, “LXRβ Reporter Cells” must be maintained at -80°C until just prior to use.
