PTEN ELISA Assay

Service Description

Provider: Echelon Biosciences was formed from a project under the Center for Cell Signaling, a State of Utah Center of Excellence, by Dr’s Glenn Prestwich and Dale Poulter (professors at the University of Utah). Since 1997, Echelon Biosciences has been offering cutting edge lipid research and lipid-protein interactions tools that are essential in elucidating lipids as top level regulators in numerous cell signaling cascades.

Background: PTEN (Phosphatase and Tensin Homolog deleted on Chromosome 10) is a phosphoinositide 3 phosphatase that converts PIP3 to PIP2 thus opposing PKB/Akt activation by PI3-K. PTEN is involved in neuronal stem cell proliferation and self-renewal, cardiac myocyte hypertrophy and contractility, and a wide range of developmental processes. PTEN, however, is best known for its role as a tumor suppressor. Loss of PTEN activity results in accumulation of PIP3, abnormal activation of PKB/Akt, unregulated cell growth, suppression of apoptosis and increased tumorigenesis in a number of human tissues. It has also been proposed that PTEN is a candidate for targeted chemotherapy because certain anti-cancer agents preferentially destroy tumors with PTEN mutations. In addition to this direct role in cancer, PTEN has recently been shown to regulate cancer-associated pathways including VEGF-mediated angiogenesis and others.

Service Details: Echelon’s PTEN ELISA is designed to detect and quantify PTEN phosphatase activity by means of a standard ELISA format, eliminating the need for radioactivity, organic solvents, and thin layer chromatography. The Echelon PTEN activity ELISA directly detects the phosphoinositide product, unlike other assays that detect free phosphate. This eliminates many possible sources of error because inorganic phosphate is the product of many phosphatase enzyme activities, and is found in common buffers and cleaning products. In addition, the PTEN activity ELISA is approximately 100 times more sensitive than free-phosphate assays and can be used with recombinant enzyme or immunoprecipitated enzyme; and potentially, cell lysate or tissue homogenate.

The PTEN Activity assay service detects PTEN phosphatase activity once the enzyme has converted PIP3 (bound to the PTEN substrate plate) to PIP2 . After the PTEN reactions are complete the PIP2 Detector and HRP conjugate are added step-wise. Colormetric detection is used to quantify the amount of PIP2 produced by PTEN phophatase activity, compared to a standard curve.

Deliverable: A downloadable data report containing raw and analyzed data including a five point standard curve. Each sample is run in triplicate and the data are expressed as mean +/- CV.

Price: The price of the service ($950) is for a minimum order of 10 samples. Bulk discounts are available according to the following table.

Sample Submission: Enter sample submission information and select service options in the Service Order Form that you can access once you add this item to your cart. Customer may submit samples containing recombinant or immunoprecipitated PTEN. Please send > 350 microliters (µL) of sample on dry ice. Upon receipt all samples will be stored at -20°C.
